Essentially, what you need to know, the short version, is the year - 2001. The events in the game are post CVX, post Umbrella Chronicles, pre-RE4. Wesker's appearance is completely healed after the fight with Chris at the end of CVX, and due to the regenerative abilities of the experimental T-Virus he injected himself with just before the mansion incident, he has aged unusually well.

Umbrella has fallen, and Wesker is currently working to rebuild it into something of his own, at the present time.

Most anything else you need to know can probably be found on wikipedia.
